Due to their many advantages such as low immunogenicity, high penetrability, engineering modification, and the ability to load different types of drugs such as nucleic acids and proteins, exosomes have been rated as a new generation drug delivery platform. Engineered exosomes are becoming important carriers of small nucleic acid molecules. In order to achieve the purpose of small nucleic acid delivery based on exosomes, different technical routes such as endogenous expression, electroporation and exogenous loading based on chemical transfection are currently available. The ExoLoad ® kit developed by Echo biotech efficiently introduces small nucleic acids such as siRNA, microRNA, and ASO into exosomes by custom-developing an exclusive transit peptide ETP (Exosome-Transit Peptide) for exosomes. It can easily and quickly achieve efficient loading of small nucleic acids in exosomes, and can be used for functional research on small nucleic acid delivery and drug treatment research.

Q: What are the storage conditions and stability of loaded exosomes? A: It is best to use it within 12 hours after loading. IF it cannot be reached, it is recommended to place it at 4°C for 3 days and not more than 7 days. Q: The shelf life of the kit and the shelf life after the transit peptide is dissolved? A: The kit has a shelf life of one year. It is recommended to use it within the validity period. The transit peptide is stable for one year at 4°C after dissolution. Q: What are the recommended operating methods for use with fat anchors? A: It is recommended to perform lipid anchoring first and then ExoLoad ®. Q: Is ExoLoad ® loaded on the surface of exosomes or inside? A: There are both on the surface and inside. You can refer to this literature. The literature shows that about half is located inside exosomes, and RNase treatment can degrade more than 40% of surface nucleic acids: Tong L, Wang Q, Zhang Y, et al. A: The ExoLoad ® small nucleic acid loading kit is specially used to load small nucleic acids of 18-30nt in exosomes. We are developing related kits for exogenous loading of long-chain mRNA, and the current performance is good. We have not tested more lncRNA or circRNA loadings at present, and the effect may not be good. Teachers who are interested can test it by themselves. The operation is easy and the results can be obtained quickly, and trial and error can be quickly made. Q: Storage temperature and transportation temperature? A: It is transported in an ice pack. It can be stored at 4℃ for a short period of time. IF it is a long period of time, it can be stored according to the conditions recommended in the instructions. A: First of all, it is certain that the feedback from our different customers is very good, and there are many types of differences in the sources of their exosomes. Secondly, it is undeniable that the loading efficiency of exosomes from different sources will be different, just like loading different small nucleic acids, the loading efficiency will not be exactly the same. Thirdly, in principle, although exosomes from different sources have a double-layer membrane structure and have slightly different charges, they are all negatively charged, so the loading efficiency will be different, but ExoLoad ® can still perform the load function. Finally, based on the same principle of loading, some articles have compared the loading differences of MSC exosomes, urine exosomes and bovine serum exosomes. The qPCR detection results are quite different, but they can all load miRNA into exosomes. Q: What is the recovery rate of the kit? A: Our formal packages (ELSR-06-02, ELSR-06-03) will come with Millipore's 100KD ultrafiltration tubes, which are recovered by ultrafiltration. The recovery rate will vary depending on the purity of the exosomes. Our recovery rate is basically maintained at 30-40% each time.
