There is no need for ultra-high-speed centrifugation equipment, which solves the problems of traditional ultra-centrifugation methods such as cumbersome steps, reliance on ultra-high-speed centrifuge equipment, difficulty in making reservations, and time and effort.

Applications
This product is suitable for serum-free cultured cell-culture supernatant, as well as large-volume samples such as urine, cerebrospinal fluid, amniotic fluid, etc. It is suitable for sample types with relatively low exosome concentration and relatively simple components for rapid exosome isolation and purification. IF the exosome concentration is particularly low and you want to increase the yield, you can first utilize a 100KDa ultrafiltration tube to concentrate the supernatant 3-5 times after the above treatment, and then proceed to the subsequent steps.

Performance
The supernatant exosomes of 293F cells isolated by ESS050 and cultured without serum were applicable to electron microscopy observation. The results are as shown above.

1. IF fetal calf serum is added to the cell-culture supernatant, can this product still be used to isolate exosomes? IF a high proportion of fetal bovine serum, such as 10% FBS, is added to the cell-culture supernatant, this product cannot effectively remove the rich lipoproteins and other impurities, and it is necessary to combine ultraviolet separation, exclusion chromatography (Exosupur® ES9P11e/ES9P14e) and other methods to further remove impurities. Utilize ESS050 to separate exosomes from the supernatant of cancer cells cultured with 10% conventional FBS fetal bovine serum for electron microscopy observation. As shown above, there are many impurities in the serum, so it is not recommended to utilize this product alone to isolate and purify exosomes from serum-containing systems. 2. How to preprocess samples? ① Collect the cell-culture supernatant, centrifuge at 300g and 4°C for 10 minutes to remove dead cells, centrifuge at 3000g and 4°C for 10 minutes to remove cell debris, centrifuge at 10000g and 4°C for 10 minutes to remove impurity debris until there is no obvious precipitation, and filter with a 0.22 μm microporous membrane. ② After the urine is fully vortexed for 1 minute, centrifuge at 3000g for 10 minutes to remove the sediment supernatant and filter with a 0.22 μm microporous membrane. The filtrate is concentrated and applicable to later utilize. This product is suitable for isolation and purification of exosomes from serum-free cultured cell-culture supernatant, urine, cerebrospinal fluid, amniotic fluid, etc., which are sample types with relatively low exosome concentration and relatively simple components. IF the exosome concentration is extremely low and you want to increase the yield, you can first utilize a 100KDa ultrafiltration tube to concentrate the supernatant 3-5 times after the above treatment, and then proceed to the subsequent steps. For other types of sample preprocessing methods, please refer to the literature or contact us. 3. The centrifugal force of the centrifuge cannot reach 10,000g? It is recommended that the centrifugal force can be reduced to 6000g and centrifuged at 4°C for 60 minutes. The reduction of time or centrifugal force will have a certain impact on the recovery rate, which can be evaluated based on your own sample conditions. 4. After adding solution A and solution B and mixing evenly, how long should it be incubated at 4°C for the longest time? It is recommended to start from 12 hours, but it is not recommended to exceed 24 hours. 5. There is no precipitation after overnight incubation and centrifugation. Is this normal? Since the exosome content in the supernatant of some cells is relatively low, the precipitate cannot be observed with the naked eye after adding liquid A and liquid B for centrifugation. It is recommended that: a. Before centrifugation, mark the approximate location of the exosomes after centrifugation to reduce losses when discarding the supernatant later. b. After centrifugation, utilize PBS to carefully pipe around the walls and bottom of the centrifuge tube during resuspension; the extracted exosomes should be quantitatively tested for NTA/NanoFCM particle concentration or BCA protein before deciding whether to conduct subsequent experiments.
