Sterility Testing
Two options are provided: rapid sterility screening and direct inoculation sterility testing (pharmacopeia-recommended). Rapid sterility screening: Method: plate spreading with visual inspection. Principle: Microbial growth on the culture medium is observed directly to quickly judge whether the sample is contaminated by aerobic microorganisms.

Sterility Testing

Direct Inoculation Sterility Method
Instrument: HTY-1800G4 sterility isolator and SPX-250B-ZII incubator. Principle: The sample is directly inoculated into liquid medium and incubated for 14 days. Turbidity or colony growth is then checked to determine contamination. This approach is more rigorous and can cover both aerobic and anaerobic organisms.
Mycoplasma Testing
Instrument: T30D gene amplification instrument. Principle: A one-step rapid mycoplasma detection kit amplifies conserved mycoplasma DNA sequences when contamination is present. The reaction color changes visibly, enabling fast screening for eight common mycoplasma species encountered during cell culture.
Endotoxin Testing
Method: Gel-clot assay (pharmacopeia-recommended). Principle: Bacterial endotoxin activates the limulus reagent cascade and ultimately forms a gel clot. The coagulation response is used to determine whether endotoxin exceeds the specified limit.
pH Measurement
Instrument: pH meter (potentiometric method, pharmacopeia-recommended). Principle: The pH of the solution is measured using a glass electrode and a reference electrode after calibration with standard buffers.
Osmolality Measurement
Method: Freezing-point depression method (pharmacopeia-recommended) using a freezing-point osmometer. Principle: Osmolality is determined indirectly from the freezing-point depression of the solution.
Insoluble Particulate Matter
Method: Light-obscuration method (pharmacopeia-recommended) using a particle analyzer. Principle: Particles passing through the sensing channel partially block incident light, generating signal changes proportional to particle cross-sectional area. This enables counting of particles within the target size range.
