The fluorescent dye in the FluoExo ® exosome tracking staining kit is a lipophilic fluorescent dye with a long aliphatic carbon tail. It can be stably integrated into the lipid region of the exosome membrane and emits green fluorescence. Its fluorescence half-life in vivo is 10-12 days. The dilution buffer in this kit is isotonic to mammalian cells and does not contain detergents or organic solvents. It is designed to maintain exosome activity during the dye labeling step while improving the solubility and staining efficiency of the dye. The pocket column is used to quickly and effectively remove the free fluorescence during the exosome labeling process to obtain purified fluorescently labeled exosomes, which can be applicable to exosome tracing studies.

1. How long can the prepared fluorescently labeled exosome final product be stored? It is recommended to utilize it as soon as possible and store it at 4℃ for no more than 48 hours. 2. Is it possible not to remove free dye? It is recommended to remove it according to the above steps to avoid interference of free dye on later experiments. 3. Will dye labeling and purification lead to loss of exosomes? There will be loss. The general recovery rate is about 30%-50% or more. The lower the concentration, the greater the loss. Therefore, it is recommended to utilize at least 2 times the exosome dose required for cell co-incubation experiments for dye labeling experiments. 4. For in vitro tracing experiments, should normal fetal bovine serum or exosome-free serum be applicable to cell culture? It is recommended to utilize exosome-free serum (Echo biotech Product No. EDFBS50) to improve the uptake efficiency of dye-labeled exosomes by cells.
