Biological tissues exhibit weaker autofluorescence in the red and near-infrared region, so red or near-infrared dyes are more suitable for in vivo exosome tracing. Near-infrared dyes are especially well suited for live-animal imaging, deep-tissue imaging, and in vivo exosome tracing. Because longer wavelengths penetrate tissue more deeply and reduce scattering and absorption, they significantly improve signal-to-noise ratio, tissue penetration depth, and imaging contrast.
The fluorescent dye in this kit is a near-infrared (NIR) cyanine dye. This kit labels the exosome surface through a bioorthogonal reaction. Unlike conventional lipophilic dyes, it is less likely to form dye aggregates that can cause false-positive signals, making it suitable for live-animal imaging, deep-tissue imaging, and in vivo exosome tracing.
The mini column included in the kit rapidly and effectively removes free reagents and fluorescent dye generated during exosome labeling, yielding purified fluorescently labeled exosomes for tracing studies.

Application Example
Using 293F exosome reference material at 3E+11 particles/mL (CTE-06), the thawed sample was labeled with an NIR dye through the bioorthogonal workflow described above, and the labeled exosomes were analyzed by nano-flow cytometry. Mice were injected via the tail vein with 200 μL, and in vivo fluorescence imaging plus ex vivo organ fluorescence detection were performed 24 h later, as shown below.
