Through transmission electron microscopy (TEM) observation, the morphological structure and size of individual exosomes can be directly observed under high magnification to identify the shape and size of exosomes. At the same time, the purity of exosomes can also be evaluated based on the cleanliness of the background, which is the gold standard for exosome characterization.
Nanoflow cytometry detection technology (NanoFCM) is a nanoscale flow cytometry technology developed on the basis of cell flow cytometry. It can directly detect exosomes at the single-vesicle level without labeling, and can detect scattered light signals of exosomes as low as 30 nm; the fluorescence channel can detect single phycoerythrin molecules and completely distinguish them from the background. The particle size and concentration information of exosomes can be directly obtained through data analysis, and the fluorescence signal of endogenous or exogenous markers of individual exosome particles can be detected.
WB is a classic experimental method for studying protein expression. Based on the biological generation method of exosomes and preliminary research, exosomes have some highly recognized biomarker proteins. By comparing the expression differences of biomarker proteins and negative proteins between mixed cells and exosomes, it was confirmed that the isolated vesicles highly express exosome biomarker proteins.

Results
The main purpose of exosome identification is to prove that the isolated high-purity vesicles are exosomes and meet the general characteristics of exosomes, which is an indispensable part of the current publication of exosome articles. Electron microscopy is the gold standard for exosome characterization, showing whether the morphological structure and size characteristics of the vesicles meet the characteristics of exosomes; size analysis mainly demonstrates whether the size of the isolated vesicles is within the size range of exosomes; WB mainly characterizes whether exosomes express known exosome protein markers; WB detection results rely on prior knowledge. The protein expression characteristics of exosomes from different species and cell lines are different. Therefore, currently only exosomes from human serum/plasma can most likely meet the requirements of three yang and one yin. It is difficult for other samples to produce three yang and one yin results due to the heterogeneity of the samples.
