Applications

Standards or controls for exosome characterization. After testing, as long as there is no handicap or there is a problem with the experimental SOP, using ContrExo® exosome standard CTE-062 for electron microscopy, particle size and WB characterization will produce a better result. Therefore, ContrExo® CTE-062 exosome standard can be used as a positive ginseng for exosome characterization experiments to conduct quality control of the experimental process. It is not limited to electron microscopy, particle size, WB three yang and one yin characterization. Other indicators such as Yang Ginseng for CD9/CD81/CD63 positive rate testing, Yang Ginseng for HLPC-SEC purity testing, and Yang Ginseng for membrane integrity testing can be used.

Advantages

1) Worry-free purchase in stock, ready to use: There is no need for tedious and time-consuming cell culture, no need to think about how to separate large-volume supernatant-derived exosomes, no need to spend time and trouble to perform large-volume supernatant separation and purification experiments of several liters or more, and no need to worry about how to perform exosome quality control. 2) Standard 9 quality controls to achieve "touchstone" quality: appearance, pH, sterility testing, particle concentration, particle size distribution, total protein concentration, purity, TEM morphology detection, exosome protein marker WB detection; Serial number Parameter item Parameter results 1 Appearance Clear liquid 2 PH 6.8-7.4 3 Sterility testing Negative 4 Particle concentration ≥3E+11 Particles/ml 5 Particle size distribution 30-150 nm 6 Total protein concentration ≤10.0 µg/µL(BCA) 7 Purity ≥5E+11 Particles/mg 8 Shape detection Clear vesicle structure (TEM) 9 Exosome protein markers 3 Yang and 1 Yin WBB) 3) Ultra-high purity: Through TFF+ density gradient centrifugation, the purity (Particles/Protein ratio) is guaranteed to be ≥5E+11 Particles/mg. 4) High safety: Full-process quality control from cell culture, supernatant harvest to exosome purification. Whole-process production environment: C+A; serum-free culture system (excluding animal-derived components); passed safety and quality control such as sterility.

FAQ

Q: Will each batch of products undergo standard characterization? A: Each batch of products will undergo standard identification, but customers need to make a separate set of characterization results when publishing articles. Q: Is this product frozen or dry powder? A: This product is currently only available in frozen form and shipped on dry ice. Please store it at -80°C after receiving it. Q: Is there anything special about this product that needs to be noted? A: First of all, our exosomes are added with pharmaceutical excipient formula buffer for storage, so that the exosomes can exist more stably. In addition, due to the characteristics of exosomes, the number of particles will decrease as the freezing time is extended. Our current test data shows that the number of particles decreases by 10-30% after 1 year of storage at -80°C. Please use the product as soon as possible after receiving the product. In addition, it is recommended to avoid repeated freezing and thawing of all exosomes. Q: Will there be any impact after adding pharmaceutical excipient formula buffer? A: Our pharmaceutical excipient formula buffer is a protective agent for exosomes, which can improve the stability of exosome storage and has no impact on the exosomes themselves, but will affect the total protein concentration detection (BCA). It is recommended to use particle number as a quantitative method, or directly use the factory protein concentration results. IF re-quantification is required, the sample needs to be replaced with a conventional laboratory buffer such as phosphate buffered saline (PBS), and then the protein concentration is measured. This method has a relatively high loss rate and is not recommended. Q: One CTE-062 exosome standard is only 100μl. Can it meet the needs of electron microscopy, particle size and WB three positive and one negative characterization experiments? A: Yes. Despite its small size and low BCA concentration (around 0.1μg/ml), CTE-062 has a high particle concentration (≥3E+11 Particles/ml) and high purity. The higher the purity, the lower the impurity protein content. Generally, WB is the most expensive method for basic characterization of samples. It is recommended that you load 10-15 μl of CTE-062 exosome standard for each protein for WB experiments.
