The concentrated product can be subjected to the Exosupur ® kit or ultra-high-speed centrifugation based on the principle of molecular size exclusion chromatography SECC) to further separate and purify exosomes, thereby improving the throughput and purity of sample processing.

For larger volumes, such as 500mL-2L, cell-culture supernatant samples are pre-concentrated, followed by Exosupur ® exclusion column or ultraviolet purification of small extracellular vesicles sEV.

Performance

When used with Exosupur ®, it can effectively recover small extracellular vesicles sEV and remove impurity proteins from large-volume samples. The larger the volume, the improved the overall performance. This kit has no effect on the morphological characteristics of exosomes.

Q: Are exosomes more difficult to dissolve when resuspended? A: This situation may be caused by the high concentration of the cell-culture supernatant and the excessive concentration factor. It is recommended to add a small volume, such as 0.2ml, and pipet repeatedly to try to dissolve. After dissolution, add to the required volume and pipet to mix. IF there is still incomplete dissolution, it is recommended to let it stand at room temperature for 30 minutes and then resuspend again. To increase the yield, mix as much as possible. IF there is still a large amount of precipitate in the supernatant after the above operation, centrifuge several times until there is no obvious precipitate. Take the centrifuged supernatant into a new centrifuge tube each time. Q: The centrifugal force of the centrifuge cannot reach 10000g? A: It is recommended that the centrifugal force can be reduced to 6000g and centrifuged at 4°C for 60 minutes. The reduction of time or centrifugal force will have a certain impact on the recovery rate, which can be evaluated based on your own sample conditions. Q: can the sample concentration be only 30-50 times? Are there any specific recommendations for concentration ratios? A: The reason why it is not recommended to utilize a concentration factor that is too high is that it is difficult to dissolve when resuspended. In addition, the exclusion column may be clogged during subsequent Exosupur ® separation, resulting in a slow flow rate. IF the cell-culture supernatant is dilute (the concentration of particles or BCA is low), the concentration factor can be increased. It is recommended that the particle concentration be >5E+10particles/ml during Exosupur ® separation to increase the exclusion yield. IF you want to simplify the operation as much as possible, according to our testing experience, you can directly concentrate it by 30 times. Q: can the concentrated sample be processed directly according to the instructions of Exosupur ®? A: The exosome purification part is carried out completely according to the instructions of Exosupur ®. It is recommended that the purified exosomes be washed twice with PBS during ultrafiltration and concentration. That is, the fraction collected by Exosupur ® is transferred to an ultrafiltration tube and first concentrated to the required volume, and then added with PBS to 4 ml. The exosomes are pipetted for cleaning, and centrifuged at 4000g. The cleaning can be repeated 2 times, and finally centrifuged to the required volume (the centrifugation time varies according to the sample concentration, generally 5-10 minutes).
