Applications

1. Remove free dye from stained exosomes; 2. Remove free fluorescent antibodies from exosomes labeled with fluorescent antibodies; 3. Removal of free small molecules after exosomes are loaded with small molecules; 4. Further purify the EV crude material with very low purity to improve the purity.

Advantages

1. Easy to operate: no special equipment is required, exosomes can be separated by direct incubation and centrifugation, and the hands-on time is 15 minutes; 2. High particle recovery rate: the secondary recovery efficiency can recover more than 60% of exosome particles by collecting only the E1 component; 3. High impurity removal efficiency: the removal efficiency of free dyes, fluorescent antibodies, small molecules, etc. exceeds 90%; 4. No dilution effect on the sample: only E1 samples are collected, which will not cause dilution to the sample; 5. High cost performance: the purification column can be cleaned and regenerated and reused 5 times.

FAQ

Q: What is the difference between this product and other Exosupur ® separation kits? A: This product has been upgraded in principle and is more suitable for the separation of trace samples. The processed sample volume is 0.1~0.5ml. It is mainly applicable to the secondary purification of exosomes. Other SEC-based Exosupur ® kits are mainly applicable to the separation of large-volume samples, processing sample volumes ranging from 0.2ml to 10ml. Q: How to choose the sample loading amount? A: The separation parameters for different separation purposes and different sample volumes are shown in Figure 1-4. You can choose higher yield or higher impurity removal rate according to the purpose of each experiment. For example, during secondary purification, loading 50μl-500μl can achieve a yield of more than 50% (ultrafiltration is generally 30%) and an impurity removal rate of more than 90%; Q: What precautions should be taken during operation? A: First try to keep the filling in the tube in a moist state. Add liquid immediately after centrifugation. IF it is not regenerated immediately, add buffer first and tighten the lower cap and screw cap for temporary storage. Secondly, IF you accidentally get the packing on the tube cover before receiving the spin column or during the operation, you can add PBS to clean and centrifuge the tube. Q: How to prepare the cleaning solution for the pocket column: 1M NaOH 30% isopropyl alcohol? A: 1M NaOH 30% isopropyl alcohol means that the final concentration of NaOH is 1M and the final concentration of isopropyl alcohol is 30% (according to volume ratio). Taking the preparation of 1L of 1M NaOH 30% isopropyl alcohol as an example, you can take 40.01g of solid NaOH + 300ml of isopropyl alcohol, dilute to 1L with pure water, and filter with a 0.22um filter membrane after preparation. IF there is no NaOH solid, taking 5M NaOH as an example, configure 100ml of 5M NaOH 20ml + 30ml isopropyl alcohol + 50ml pure water, mix and filter with a 0.22um filter membrane. Q: Is it necessary to shake the sample during incubation? What special equipment is needed to shake it up? A: It is recommended to rotate and incubate to make the sample and filler contact more fully and achieve improved separation effect. No special equipment is required, just a shaker used in the laboratory, shake at 80 rpm for 30 minutes. Q: Will it have any impact IF it enters the tube cover during rotation and incubation? A: There is no impact. After incubation for 30 minutes, it will all enter the Exosupur ® pocket column once centrifuged. Q: How to store the isolated exosomes? A: The problem of exosome preservation has nothing to do with the isolation method, but is determined by its own characteristics. IF conditions permit, it is best to utilize fresh exosomes for experiments. Frozen exosomes are prone to agglomeration and precipitation. In actual research, exosome functional experiments often require a large amount of exosomes that have to be extracted and stored in batches and can be stored at -80°C; however, when doing biomarker research, such as RNAomics, follow-up experiments such as RNA extraction must be completed as soon as possible after exosomes are extracted. Q: What type of pocket column for exosomes can improve the quality of electron microscopy? A: Under the electron microscope, there is obviously a layer of impurities covering the surface, but the vesicles can be vaguely seen, as shown in the picture below. You can consider using a pocket column for secondary purification and take another photo to improve the quality of the electron microscope. IF there are only impurities and invisible vesicles, there is a certain probability that there are only impurities, so it is not recommended to try.
